    RNA-inspired intramolecular transesterification accelerates the hydrolysis of polyethylene-like polyphosphoesters

    To synthesize new (bio)degradable alternatives to commodity polymers, adapting natural motives can be a promising approach. We present the synthesis and characterization of degradable polyethylene (PE)-like polyphosphoesters, which exhibit increased degradation rates due to an intra-molecular transesterification similar to RNA. An α,ω-diene monomer was synthesized in three steps starting from readily available compounds. By acyclic diene metathesis (ADMET) polymerization, PE-like polymers with molecular weights up to 38 400 g mol(−1) were obtained. Post-polymerization functionalization gave fully saturated and semicrystalline polymers with a precise spacing of 20 CH(2) groups between each phosphate group carrying an ethoxy hydroxyl side chain. This side chain was capable of intramolecular transesterification with the main-chain similar to RNA-hydrolysis, mimicking the 2â€Č-OH group of ribose. Thermal properties were characterized by differential scanning calorimetry (DSC (T(m)ca. 85 °C)) and the crystal structure was investigated by wide-angle X-ray scattering (WAXS). Polymer films immersed in aqueous solutions at different pH values proved an accelerated degradation compared to structurally similar polyphosphoesters without pendant ethoxy hydroxyl groups. Polymer degradation proceeded also in artificial seawater (pH = 8), while the polymer was stable at physiological pH of 7.4. The degradation mechanism followed the intra-molecular “RNA-inspired” transesterification which was detected by NMR spectroscopy as well as by monitoring the hydrolysis of a polymer blend of a polyphosphoester without pendant OH-group and the RNA-inspired polymer, proving selective hydrolysis of the latter. This mechanism has been further supported by the DFT calculations. The “RNA-inspired” degradation of polymers could play an important part in accelerating the hydrolysis of polymers and plastics in natural environments, e.g. seawater

    Incrementally Verifiable Computation via Incremental PCPs

    If I commission a long computation, how can I check that the result is correct without re-doing the computation myself? This is the question that efficient verifiable computation deals with. In this work, we address the issue of verifying the computation as it unfolds. That is, at any intermediate point in the computation, I would like to see a proof that the current state is correct. Ideally, these proofs should be short, non-interactive, and easy to verify. In addition, the proof at each step should be generated efficiently by updating the previous proof, without recomputing the entire proof from scratch. This notion, known as incrementally verifiable computation, was introduced by Valiant [TCC 08] about a decade ago. Existing solutions follow the approach of recursive proof composition and can be based on strong and non-falsifiable cryptographic assumptions (so-called ``knowledge assumptions\u27\u27). In this work, we present a new framework for constructing incrementally verifiable computation schemes in both the publicly verifiable and designated-verifier settings. Our designated-verifier scheme is based on somewhat homomorphic encryption (which can be based on Learning with Errors) and our publicly verifiable scheme is based on the notion of zero-testable homomorphic encryption, which can be constructed from ideal multi-linear maps [Paneth and Rothblum, TCC 17]. Our framework is anchored around the new notion of a probabilistically checkable proof (PCP) with incremental local updates. An incrementally updatable PCP proves the correctness of an ongoing computation, where after each computation step, the value of every symbol can be updated locally without reading any other symbol. This update results in a new PCP for the correctness of the next step in the computation. Our primary technical contribution is constructing such an incrementally updatable PCP. We show how to combine updatable PCPs with recently suggested (ordinary) verifiable computation to obtain our results

    We briefly summarize the reported anomalous effects in deuterated metals at ambient temperature, commonly known as "Cold Fusion" (CF), with an emphasis on important experiments as well as the theoretical basis for the opposition to interpreting them as cold fusion.     Early postnatal hypotension is not associated with indicators of white matter damage or cerebral palsy in extremely low gestational age newborns

    ObjectivesTo evaluate, in extremely low gestational age newborns (ELGANs), relationships between indicators of early postnatal hypotension and cranial ultrasound indicators of cerebral white matter damage imaged in the nursery and cerebral palsy diagnoses at 24 month follow-up.MethodsThe 1041 infants in this prospective study were born at < 28 weeks gestation, were assessed for 3 indicators of hypotension in the first 24 postnatal hours, had at least one set of protocol cranial ultrasound scans, and were evaluated with a structured neurologic exam at 24 months corrected age. Indicators of hypotension included: 1) lowest mean arterial pressure (MAP) in the lowest quartile for gestational age; 2) treatment with a vasopressor; and 3) blood pressure lability, defined as the upper quartile of the difference between each infant’s lowest and highest MAP. Outcomes included indicators of cerebral white matter damage, i.e. moderate/severe ventriculomegaly or an echolucent lesion on cranial ultrasound, and cerebral palsy diagnoses at 24 months gestation. Logistic regression was used to evaluate relationships among hypotension indicators and outcomes, adjusting for potential confounders.ResultsTwenty-one percent of surviving infants had a lowest blood pressure in the lowest quartile for gestational age, 24% were treated with vasopressors, and 24% had labile blood pressure. Among infants with these hypotension indicators, 10% percent developed ventriculomegaly and 7% developed an echolucent lesion. At 24-months follow-up, 6% had developed quadriparesis, 4% diparesis, and 2% hemiparesis. After adjusting for confounders, we found no association between indicators of hypotension, and indicators of cerebral white matter damage or a cerebral palsy diagnosis.ConclusionsThe absence of an association between indicators of hypotension and cerebral white matter damage and or cerebral palsy suggests that early hypotension may not be important in the pathogenesis of brain injury in ELGANs

    A new scheme to calculate isotope effects

    We present a new scheme to calculate isotope effects. Only selected frequencies at the target level of theory are calculated. The frequencies are selected by an analysis of the Hessian from a lower level of theory. We obtain accurate isotope effects without calculating the full Hessian at the target level of theory. The calculated frequencies are very accurate. The scheme converges to the correct isotope effect

    Whether weather matters: Evidence of association between in utero meteorological exposures and foetal growth among Indigenous and non-Indigenous mothers in rural Uganda

    Pregnancy and birth outcomes have been found to be sensitive to meteorological variation, yet few studies explore this relationship in sub-Saharan Africa where infant mortality rates are the highest in the world. We address this research gap by examining the association between meteorological factors and birth weight in a rural population in southwestern Uganda. Our study included hospital birth records (n = 3197) from 2012 to 2015, for which we extracted meteorological exposure data for the three trimesters preceding each birth. We used linear regression, controlling for key covariates, to estimate the timing, strength, and direction of meteorological effects on birth weight. Our results indicated that precipitation during the third trimester had a positive association with birth weight, with more frequent days of precipitation associated with higher birth weight: we observed a 3.1g (95% CI: 1.0–5.3g) increase in birth weight per additional day of exposure to rainfall over 5mm. Increases in average daily temperature during the third trimester were also associated with birth weight, with an increase of 41.8g (95% CI: 0.6–82.9g) per additional degree Celsius. When the sample was stratified by season of birth, only infants born between June and November experienced a significant associated between meteorological exposures and birth weight. The association of meteorological variation with foetal growth seemed to differ by ethnicity; effect sizes of meteorological were greater among an Indigenous subset of the population, in particular for variation in temperature. Effects in all populations in this study are higher than estimates of the African continental average, highlighting the heterogeneity in the vulnerability of infant health to meteorological variation in different contexts. Our results indicate that while there is an association between meteorological variation and birth weight, the magnitude of these associations may vary across ethnic groups with differential socioeconomic resources, with implications for interventions to reduce these gradients and offset the health impacts predicted under climate change

    No-signaling Linear PCPs

    In this paper, we give a no-signaling linear probabilistically checkable proof (PCP) system for polynomial-time deterministic computation, i.e., a PCP system for P such that (1) the honest PCP oracle is a linear function and (2) the soundness holds against any (computational) no-signaling cheating prover, who is allowed to answer each query according to a distribution that depends on the entire query set in a certain way. To the best of our knowledge, our construction is the first PCP system that satisfies these two properties simultaneously. As an application of our PCP system, we obtain a 2-message delegating computation scheme by using a known transformation. Compared with the existing 2-message delegating computation schemes that are based on standard cryptographic assumptions, our scheme requires preprocessing but has a simpler structure and makes use of different (possibly cheaper) standard cryptographic primitives, namely additive/multiplicative homomorphic encryption schemes

    Association of Severe Bronchiolitis during Infancy with Childhood Asthma Development: An Analysis of the ECHO Consortium

    Objective: Many studies have shown that severe (hospitalized) bronchiolitis during infancy is a risk factor for developing childhood asthma. However, the population subgroups at the highest risk remain unclear. Using large nationwide pediatric cohort data, namely the NIH Environmental influences on Child Health Outcomes (ECHO) Program, we aimed to quantify the longitudinal relationship of bronchiolitis hospitalization during infancy with asthma in a generalizable dataset and to examine potential heterogeneity in terms of major demographics and clinical factors. Methods: We analyzed data from infants (age <12 months) enrolled in one of the 53 prospective cohort studies in the ECHO Program during 2001–2021. The exposure was bronchiolitis hospitalization during infancy. The outcome was a diagnosis of asthma by a physician by age 12 years. We examined their longitudinal association and determined the potential effect modifications of major demographic factors. Results: The analytic cohort consisted of 11,762 infants, 10% of whom had bronchiolitis hospitalization. Overall, 15% subsequently developed asthma. In the Cox proportional hazards model adjusting for 10 patient-level factors, compared with the no-bronchiolitis hospitalization group, the bronchiolitis hospitalization group had a significantly higher rate of asthma (14% vs. 24%, HR = 2.77, 95%CI = 2.24–3.43, p < 0.001). There was significant heterogeneity by race and ethnicity (Pinteraction = 0.02). The magnitude of the association was greater in non-Hispanic White (HR = 3.77, 95%CI = 2.74–5.18, p < 0.001) and non-Hispanic Black (HR = 2.39, 95%CI = 1.60–3.56; p < 0.001) infants, compared with Hispanic infants (HR = 1.51, 95%CI = 0.77–2.95, p = 0.23). Conclusions: According to the nationwide cohort data, infants hospitalized with bronchiolitis are at a higher risk for asthma, with quantitative heterogeneity in different racial and ethnic groups
